Cultures et élevages de masse dans le contexte d'une écloserie-nurserie de mollusques bivalves : les productions phytoplanctoniques et le grossissement des post-larves ArchiMer
Le Borgne, Y.; Marin, J.; Vergonzanne, G..
The hatchery-nursery from the Société Atlantique de Mariculture settled in Barfleur studied within the ECOTRON program set up by C.N.E.X.O. the characteristics of mass cultures for monospecific phytoplancton and of the rearing of oyster spat (Ostrea edulis. Crassostrea giqas) and clam spat (Venerupis semidecussata) . The maintenance features of the cultures in order to obtain cell densities around 1 million cell/ml. (or 50 g. of dry weight/m3) in containers up to 5000 liters have been established for the main species used in aquaculture (Isochrysis qalbana. Monochrysis lutheri, Tetraselmis suecica. Skeletonema costatum. Phaeodactylum tricornutum. Cyclotella nana, Chaetoceros calcitrans). The spat growth in nursery without providing any cultured food...

Production artificielle de naissain de coquilles Saint-Jacques Pecten maximus (L.). Premiers résultats en rade de Brest ArchiMer
Buestel, Dominique; Cochard, Jean-claude; Dao, Jean-claude; Gerard, Andre.
Artificial production from natural spawners could be a way of getting big quantities of spat of Pecten maximus, ln the method described, larval phase (until a size of 250 microns) occurs in a hatchery, post-Iarval phase (from 250 µ until 3 millimeters) in a nursery, and first growth period directly into the sea (from 2 mm to a size of 25-30 mm), The fifst results obtained, production of several thousand of individuals good enough for reseeding, seem to be transposable to a larger scale.

Mortalités massives en élevage larvaire de Lates Calcarifer ( Mise en évidence d'un virus neurotrope ) ArchiMer
Renault, Tristan; Thouard, Emmanuel; Weppe, Maurice.
Since 1986 high mortality of hatchery-reared sea bass, Lates Calcarifer, larvae appeared in Ifremer's Pacifie Oceanological Center (Tahiti). Light and electron microscopy show vacuolation and cells with cytoplasmic inclusion bodies in the brain, the cord and the eye relina of affected larvae. Numerous non-enveloped particles identified as viral units, icosahedral in morphology and measuring about 25 nm in diameter were found in the cytoplasm of the degenerating cells. In affected larvae, these particles were isolated by density grandient centrifugations. In vitro tests on cell lines and in vivo test on fish are performed with samples of diseased larvae. And immunisation of mice are made with purified immunoglobulins of Lates calcarifer.

Etudes sur les mortalites larvaires de la coquille st Jacques Pecten maximus en ecloserie ArchiMer
Robert, Rene; Miner, Philippe; Nicolas, Jean-louis; Mazuret, Michel; Connan, Jean-paul.
Larval mortalities occurring in molluscan hatcheries have often been associated with bacterial contamination. Although batches of oyster and clam larvae have been routinely reared in the hatchery of Argenton without antibiotics, high larval mortalities have been recorded with Pecten maxirnus under similar conditions. For this species, an addition of chloramphenicol was found necessary. However, this chemical has now been banned in Europe. Thus either substitition products or an improvement in the rearing procedures is essential. Studies carried out have shown that neither a decrease in larval density (1 larvae. ml"l) nor sea water change frequency (1 per day) had any positive effects. Furthermore, elective substances such as sugars were not suitable and...

Substitutes for live microalgae in mariculture: a review ArchiMer
Robert, Rene; Trintignac, P.
In mollusc hatcheries and nurseries, live microalgae are traditionally used as feed for bivalves. Despite recent improvements in efficiency, such intensive culture of microalgae represents major cost factors in the production of bivalve larvae and spat. In an attempt to provide cost-effective alternatives and to simplify hatchery-nursery procedures, non-living algal food has been developed over the past few years. Substitution products such as bacteria, yeast, vitamin or docosahexaenoic acid enriched powders, dry and concentrated microalgae and microcapsules have been tested and evaluated on an experimental scale. The present article reviews major research aimed at developing non living diets in mariculture, which generally resulted in lower growth and...

Le système immunitaire du bar Dicentrarchus labrax. Immunoglobulines, cellules productrices d'immunoglobulines, ontogenèse applications à une pathologie virale néonatale ArchiMer
Breuil, Gilles.
The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. On adult basses, these immunoglobulins are tetrameric molecules with high molecular weight (850 kD), made of heavy (H) - 80 kD and light (L) - 26 kD chains. In the eggs, simplest forms such as monomeric or dimeric are brought to the fore; they are located under...

Bivalve hatchery technology: The current situation for the Pacific oyster Crassostrea gigas and the scallop Pecten maximus in France ArchiMer
Robert, Rene; Gerard, Andre.
The French oyster farming industry relies almost exclusively on juveniles collected from the natural environment; the supply of spat produced by hatcheries is low, about 10% of the industry's requirements. Development through selective breeding of oyster stocks which are better suited for aquaculture purposes, is likely to reverse this tendency since only hatcheries will be able to supply such animals. Scallop farming, which at present is poorly developed in France, relies exclusively on hatchery produced spat. Although hatchery technology is constantly being improved, significant production problems remain which must be solved before hatcheries become a major supplier of juveniles for the industry. This paper describes the present state of hatchery...

The role of microalgae in aquaculture: situation and trends ArchiMer
Muller-feuga, Arnaud.
Algae are utilized diversely in aquaculture, but their main applications are related to nutrition. They are used in toto, as a sole component or as a food additive to supply basic nutrients, color the flesh of salmonids or for other biological activities. The need for nutritional sources safer than traditional animal products has renewed interest in plants in general and algae in particular. This report deals principally with the nutritional role of microalgae in aquaculture. The larvae of molluscs, echinoderms and crustaceans as well as the live prey of some fish larvae feed on microalgae. Though attempts have been made to substitute inert particles for these micro-organisms which are difficult to produce, concentrate and store, only shrimp and live prey...

Microsatellite markers as a tool to study reproductive success in the Pacific oyster, Crassostrea gigas (Thunberg), under controlled hatchery conditions ArchiMer
Boudry, Pierre; Collet, Bertrand; Cornette, Florence; Hervouet, Veronique; Bonhomme, Francois.
Oysters, like many marine species have a very high fecundity. Previous studies have shown that populations, from both hatcheries and the natural environment, have very low Ne/N ratios. These observations reveal high variation in reproductive success. In order to study individual reproductive success under controlled conditions, we used microsatellite markers to quantify parental contributions in in vitro crosses (5 males and 5 females) of Crassostrea gigas, the Pacific oyster. High polymorphism of the microsatellites (more that 50 alleles per locus) eased the parentage identifications. The results of a cross allowing gametic competition were compared with the results from a second cross where the gametes of the same parents were kept separate for each...

Recherche et identification de bactéries pathogènes au sein d'une écloserie nurserie d'huître creuse, Crassostrea gigas, en relation avec la mortalité estivale du naissain ArchiMer
Waechter, Magali.
Summer mortality has been observed on oyster spat Crassostrea gigas since 1991 on french coasts. In our study, we deseribed symptoms observed during the mortality, especially adductor muscle weakness and we isolated strains on Zobell medium and TCBS from moribund spat. Strains isolated during a high mortality (60 %) were selected by experimental pathology on spat. Inoculation by bath led us to select a pathogenic strain (TNEMF6) for oyster spat. Phenotypic identification, phylogenetic study on 16S rDNA and quantitative DNA/DNA hybridisation showed that this strain belongs to Vibrio splendidus biovar II. Phenotyping and ribotyping with strains isolated during the study showed that only TNNIII7 and TNMB5 are similar to the pathogenic strain TNEMF6....

L'élevage larvaire de crevettes en Nouvelle-Calédonie. L'Oxytétracycline et l'association Triméthoprime - Sulfadiazine en tant qu'alternatives à l'emploi de l'Erythromycine en écloserie ArchiMer
Francois, Cyrille.
Aquaculture of penaeid shrimps holds a major place in productions of animals. New Caledonia takes part to these productions by developing the breeding of Pacific blue shrimp Litopenaeus stylirostris. Nowadays, penaeid larviculture can’t be undertaken unless antibiotics are used. Experiments in the Caledonian Aquacultural Laboratory (LAC IFREMER Saint-Vincent) put across to the development of two antibiotic prophylaxis plans for substitution of erythromycine, one plan with oxytetracycline (hydrochloride), the other plan with trimethoprime-sulfadiazine association. After an antimicrobial agent susceptibility testing of bacteria and an acute toxicity study, efficacity assays are the biological validation of these projects of protocols

A comparative study of the reproductive cycle of Ruditapes decussatus under natural (South of Tunisia) and controlled conditions (hatchery). The aim of this study was to describe and compare the gametogenic activity of Ruditapes decussatus in situ and in hatchery. Criteria of maturity were characterized and the positive role of conditioning in the success of artificial reproduction was underlined. Specimens sampled in the South of Tunisia were conditioned at the experimental station of Monastir (Tunisia). The parameters studied were the variation of the condition index and the frequency distribution of oocyte diameters. Results showed that the reproductive cycle was continuous in situ. Gametogenesis started in January and February. Gonadic activity...

in mollusc hatcheries. Consequently, we assessed themost commonly used species to establish their growth and size profiles, their cytomorphological, biochemical, biomolecular identification as well as their cytometer signs, and also their ability to grow under extreme conditions of temperature and salinity. This work aimed at matching the conformity of the strains used in Argenton (dispatched to most of the French commercial hatcheries) to those of a referenced culture collection. A previous screening preceded this study, because of the 45 species and/or clones held originally, Argenton kept no more than 15 species. As most of the latter are also represented in referenced collection, our strains are therefore correctly identified. On the other hand, the...

Conséquences génétiques de la production intensive de larves d'huîtres en écloserie : étude des processus de dérive et de sélection liés aux pratiques d'élevage. ArchiMer
Taris, Nicolas.
In order to study the genetic influence of intensive production of the larval stage in commercial hatcheries, two types of specific selective processes were examined: the effects of discarding the smallest larvae and temperature. A mixed-family approach was used in order to infer the genetic composition of the larvae. The results show that our mixed-family approach, combined with microsatellite-based family assignment, is a powerful tool for the study of bivalve larvae genetics. Selective sieving is an advantageous practice at a phenotypic scale, but also represents a substantial risk for diversity loss. The culled larvae are important for the minimization of the variability in reproductive success and the maximization of broodstock genetic diversity....

Taris, Nicolas; Sauvage, Christopher; Batista, Frederico; Baron, Sophie; Ernande, Bruno; Haffray, Pierrick; Boudry, Pierre.
Previous studies have shown heritable variation in larval developmental traits in the Pacific oyster Crassostrea gigas. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors, specific to hatcheries, were examined: the effect of discarding the smallest larvae (i.e. culling) and the effect of temperature (20°C versus 26°C). A mixed-family approach was used in order to infer the genetic composition of larval populations and family assignment, limiting possible environmental bias and allowing the study of a relatively large number of families using a limited number of larval tanks. Our results show that three multiplexed highly polymorphic microsatellite markers are a powerful tool for family assignment and,...

Phenotypic and genetic consequences of size selection at the larval stage in the Pacific oyster (Crassostrea gigas) ArchiMer
Taris, Nicolas; Ernande, Bruno; Mccombie, Helen; Boudry, Pierre.
The life histories of oysters in the genus Crassostrea, like those of most marine bivalves, are typified by high fecundity and low survival in nature. Rearing conditions in hatcheries however ensure optimized density, diet, and temperature. Hatcheries are becoming increasingly important for the production of juveniles in aquaculture, and their culture practices often include culling of slow growing larvae to reduce and synchronize the time taken to reach settlement. Because previous studies have found substantial genetic variation for early life developmental traits in Crassostrea gigas, these culling practices are likely to cause highly different selective pressures in hatcheries from those in the natural environment. We studied the phenotypic and genetic...

Response to unintentional selection for faster development associated with inbreeding depression in Crassostrea gigas larvae, resulting from a broodstock selective breeding plan ArchiMer
Taris, Nicolas; Batista, Frederico; Marissal, Eric; Boudry, Pierre.
Direct and indirect consequences of selective breeding in marine bivalves still remain largely unexplored. For species with two-phase life cycle, like the Pacific oyster (Crassostrea gigas), most studies have focused on juvenile and adult stages, but relatively few have focused on juvenile and adult stages, but relatively few have considered the larval stage, especially in a domestication context. We assessed the impact of hatchery practices on larval traits, notably on larval growth (due to the culling of slow growing larvae), by the study of larval progenies. Larvae originating from crosses using parental oysters both from natural beds and from hatchery broodstock which had been selected for adult growth and shell shape for seven generations. A set of...
